ABSTRACT

ABSTRACT Introduction: The most evident change during exercise is the increase in oxygen demand, and prolonged high-intensity exercises may impact this even more on this demand. Physical training with progressive loads is a method that aims to reach the training goals by adjusting factors such as initial load, stage load, and a load of the athletes and can be a monitor of the aerobic and anaerobic exercise on the athlete with potential benefits for the high demand of oxygenation. Objective: Study the changes in heart rate and indexes of cardiorespiratory endurance of sprinters under training with progressive loads. Methods: By random sampling, the study subjects were 15 male sprinters and 15 non-sportsmen as research volunteers. The study subjects performed a protocol with progressive load training on a platform. All subjects underwent the Italian Kosmai cardiopulmonary function test before and after training. Results: The subjects who did not practice the sport showed VO2 max values at an exercise intensity of 13 km/h. The VO2 max maximum for sprinters was 15 km/h. Individuals who were not sprinters generally showed lower VT levels than sprinters (P <0.05). Average non-speeder individuals demonstrated higher MET scores than sprinters. Conclusion: Cardiopulmonary endurance and energy metabolism levels are strongly related to long-term participation in physical activity and exercise intensity. Incremental load training improved cardiorespiratory fitness in sprinters. The results may provide a specific theoretical basis for the daily training of sprinters in the future. RESUMO Introdução: A alteração mais evidente durante o exercício é o aumento da demanda de oxigênio e os exercícios prolongados de alta intensidade podem impactar ainda mais nessa demanda. O treinamento físico com cargas progressivas é um método que visa atingir as metas de treinamento ajustando fatores como carga inicial, carga de estágio e uma carga dos atletas e pode ser um monitor do exercício aeróbico e anaeróbico sobre o atleta com potenciais benefícios para a alta demanda de oxigenação. Objetivo: Estudar as alterações na frequência cardíaca e nos índices de resistência cardiorrespiratória de velocistas sob treinamento com cargas progressivas. Métodos: Selecionou-se 15 velocistas do sexo masculino e 15 homens que não praticavam o esporte como voluntários de pesquisa por amostragem aleatória. Os sujeitos do estudo realizaram um protocolo com treinamento de cargas progressivas em plataforma. Todos os indivíduos foram submetidos ao teste de função cardiopulmonar Italian Kosmai antes e após o treinamento. Resultados: Os indivíduos que não praticavam o esporte apresentaram valores de VO2max em uma intensidade de exercício de 13 km/h. O VO2max máximo para velocistas foi de 15 km/h. Indivíduos que não eram velocistas em geral apresentaram níveis de VT mais baixos do que os velocistas (P <0,05). Os não velocistas médios demonstraram pontuações MET mais altas do que os velocistas. Conclusão: A resistência cardiopulmonar e os níveis de metabolismo energético estão fortemente relacionados com a participação a longo prazo na atividade física e intensidade do exercício. O treinamento de carga incremental melhorou a aptidão cardiorrespiratória em velocistas. Os resultados podem fornecer uma base teórica específica para o treinamento diário de velocistas no futuro. RESUMEN Introducción: La alteración más evidente durante el ejercicio es el aumento de la demanda de oxígeno y los ejercicios prolongados de alta intensidad pueden incidir aún más en esta demanda. El entrenamiento físico con cargas progresivas es un método que pretende alcanzar los objetivos de entrenamiento ajustando factores como la carga inicial, la carga por etapas y la carga de los atletas y puede ser un monitor del ejercicio aeróbico y anaeróbico en el atleta con beneficios potenciales para la alta demanda de oxigenación. Objetivo: Estudiar las alteraciones en la frecuencia cardíaca y en los índices de resistencia cardiorrespiratoria de los velocistas bajo tratamiento con cargas progresivas. Métodos: Seleccionamos a 15 velocistas masculinos y a 15 hombres no practicantes como voluntarios de la investigación mediante un muestreo aleatorio. Los sujetos se sometieron a un protocolo con entrenamiento de carga progresiva en plataforma. Todos los sujetos fueron sometidos a la prueba de función cardiopulmonar de Kosmai italiano antes y después del entrenamiento. Resultados: Los sujetos que no practicaban el deporte presentaron valores de VO2máx a una intensidad de ejercicio de 13 km/h. El VO2máx de los velocistas era de 15 km/h. Los individuos que no eran velocistas en general mostraron niveles de VT más bajos que los velocistas (P <0,05). La media de los no-principiantes mostró mayores puntuaciones MET que los velocistas. Conclusión: Los niveles de resistencia cardiopulmonar y de metabolismo energético están fuertemente relacionados con la participación a largo plazo en la actividad física y la intensidad del ejercicio. El entrenamiento de carga incremental mejoró la aptitud cardiorrespiratoria de los velocistas. Los resultados pueden proporcionar una base teórica específica para el entrenamiento diario de los velocistas en el futuro. ABSTRACT Introduction Badminton has a wide audience and a large development market in China. Although it benefits the flexibility of sedentary workers by recruiting the whole body's musculature, it can also cause a harmful effect due to lack of preparation or intensity during matches. Objective Study the methods of sports injury and rehabilitation of badminton players. Methods 60 badminton players with low back injuries were randomly divided into experimental and control groups. The control group used the traditional method of recovery by manual techniques. An intervention with core stability training lasting 40 minutes three times a week for six weeks was added to the experimental group. Results The degree of injury of badminton players is mostly mild, with some cases of moderate and severe injury. The combination of core strengthening associated with manual therapy rehabilitation and stretching can accelerate the muscular rehabilitation of the lumbar spine. Conclusion In the rehabilitation process of badminton injuries, we should combine physical training with traditional rehabilitation techniques, increasing the effectiveness of sports rehabilitation. RESUMO Introdução O badminton tem um público amplo e um grande mercado de desenvolvimento na China. Embora beneficie a flexibilidade de trabalhadores sedentários ao recrutar toda a musculatura corporal, ele também pode causar um efeito deletério devido a falhas na preparação ou intensidade durante os jogos. Objetivo Estudar os métodos de lesão esportiva e reabilitação de jogadores de badminton. Métodos 60 jogadores de badminton com lesão lombar foram divididos aleatoriamente em grupo experimental e grupo controle. O grupo controle utilizou o método tradicional de recuperação por técnicas manuais. Ao grupo experimental foi adicionada uma intervenção com treinamento de estabilidade do core com duração de 40 minutos, três vezes por semana, durante seis semanas. Resultados O grau de lesão dos jogadores de badminton é majoritariamente leve, com alguns casos de lesão moderada e grave. A combinação do fortalecimento do core associada à reabilitação por terapia manual e alongamentos pode acelerar a reabilitação muscular da coluna lombar. Conclusão No processo de reabilitação de lesões de badminton, devemos combinar o treinamento físico com as técnicas de reabilitação tradicionais, aumentando a eficácia da reabilitação esportiva. RESUMEN Introducción El bádminton tiene un amplio público y un gran mercado de desarrollo en China. Aunque beneficia a la flexibilidad de los trabajadores sedentarios al reclutar toda la musculatura del cuerpo, también puede causar un efecto deletéreo debido a la falta de preparación o intensidad durante los partidos. Objetivo Estudiar los métodos de lesión deportiva y rehabilitación de los jugadores de bádminton. Métodos 60 jugadores de bádminton con lesión lumbar fueron divididos aleatoriamente en grupo experimental y grupo de control. El grupo de control utilizó el método tradicional de recuperación mediante técnicas manuales. Al grupo experimental se le añadió una intervención con entrenamiento de estabilidad del núcleo de 40 minutos, tres veces por semana, durante seis semanas. Resultados El grado de lesión de los jugadores de bádminton es mayoritariamente leve, con algunos casos de lesiones moderadas y graves. La combinación de fortalecimiento del core asociada a la rehabilitación de terapia manual y a los estiramientos puede acelerar la rehabilitación muscular de la columna lumbar. Conclusión En el proceso de rehabilitación de las lesiones de bádminton, debemos combinar el entrenamiento físico con las técnicas de rehabilitación tradicionales, aumentando la eficacia de la rehabilitación deportiva. @#[摘 要] 目的:探讨在靶向HER2的CAR的CD3ζ链胞内区引入YRHQ基序对CAR-T细胞的特异性杀伤活性及免疫记忆形成的影响。方法:通过DNA合成获得包含靶向HER2的编码抗原受体H28ζ或H28ζ(YRHQ)的DNA片段,通过慢病毒载体将不同CAR的DNA片段分别转导健康人外周血T细胞,制备靶向HER2的H28ζ-CAR-T及H28ζ(YRHQ)-CAR-T细胞。扩增过程中对不同CAR-T细胞进行计数,FCM检测CAR的表达率。将CAR-T细胞分别与HER2阳性的SKOV3、MDA-MB-453或HER2阴性的MCF-7细胞共培养,LDH释放法检测其杀伤活性,ELISA法检测IL-2、IFN-γ和颗粒酶B的水平,WB法检测STAT3磷酸化水平及免疫检查点分子TIM-3和PD-1的表达,通过FCM检测CCR7、CD45RO的表达,分析CAR-T细胞的表型。结果:H28ζ-CAR-T和H28ζ(YRHQ)-CAR-T细胞扩增能力较好,体外培养7 d时扩增4~5倍。H28ζ-CAR和H28ζ(YRHQ)-CAR表达率分别为(33.3±2.85)%和(28.30±3.2)%。H28ξ(YRHQ)-CAR-T细胞的杀伤活性较H28ζ-CAR-T细胞更高(P<0.05)。经HER2抗原刺激后,与T细胞或H28z-CAR-T细胞比较,H28ξ(YRHQ)-CAR-T细胞的STAT3磷酸化水平较H28ξ-CAR-T细胞明显升高(P<0.01);而两者间PD-1和TIM-3的表达无明显差异。未经抗原刺激的CAR-T细胞CCR7和CD45RO表达与正常T细胞比较差异无统计学意义(均P>0.05),与SKOV3细胞共培养后,与T细胞或H28z-CAR-T细胞比较,H28ξ(YRHQ)-CAR-T细胞中TEM细胞比例明显增加、TCM细胞比例明显减少(均P<0.05)。结论:在CD3胞内区引入YRHQ基序可在一定程度上提高CAR-T细胞的杀伤潜力。

@#Triclosan, widely used in various personal care products, is one of the most common environmental endocrine disruptors in our life. It can be detected from water, soil and dust, and its environmental exposure level increased with consumption. Human body are exposed to triclosan through food and water, but the evidence that triclosan exposure leads to adverse health effects is not sufficient. This paper summarizes the progress of studies related to environmental concentration, population exposure and health risks of triclosan in recent years.

@#Phthalic acid esters (PAEs) are commonly used plasticizers and solvents. Human body is exposed and absorbed mainly through diet, skin and air inhalation. The biological samples such as urine, blood, saliva, semen and breast milk generally contain PAEs and their metabolites, but the concentrations of PAEs metabolites vary in different samples. In the general population, the levels of PAEs are higher in children than in adults, and higher in women than in men; the levels of PAEs are higher in the occupational population than in the general population. In this paper, the research of PAEs related human biomonitoring in the general population and occupational population at home and abroad is reviewed, so as to provide the basis for reducing the exposure of PAEs and related health risk.

ABSTRACT

Wound scarring remains a major challenge for plastic surgeons. Transforming growth factor (TGF)-β plays a key role in the process of scar formation. Previous studies have demonstrated that truncated TGF-β type II receptor (t-TGF-βRII) is unable to continue signal transduction but is still capable of binding to TGF-β, thereby blocking the TGF-β signaling pathway. Hepatocyte growth factor (HGF) is a multifunctional growth factor that promotes tissue regeneration and wound healing. Theoretically, the combination of HGF and t-TGF-βRII would be expected to exert a synergistic effect on promoting wound healing and reducing collagen formation. In the present study, lentivirus-mediated transfection of the two genes (t-TGF-βRII/HGF) into fibroblasts in vitro and in a rat model in vivo was used. The results demonstrated that the expression of t-TGF-βRII and HGF in NIH-3T3 cells was successfully induced. The expression of both molecules significantly reduced collagen I and III expression, and also inhibited fibroblast proliferation. Furthermore, histological examination and scar quantification revealed less scarring in the experimental wound in a rat model. Moreover, on macroscopic inspection, the experimental wound exhibited less visible scarring compared with the control. Therefore, the present study demonstrated that the combination gene therapy of t-TGF-βRII and HGF promoted wound healing, with less scarring and more epithelial tissue formation, not only by suppressing the overgrowth of collagen due to its antifibrotic effect, but also by promoting tissue regeneration.

@# Objective: To evaluate the potential associations between the single nucleotide polymorphisms (SNPs) of lncRNA H19 genes and the susceptibility to gastric cancer (GC), especially to EBV-associated gastric cancer (EBVaGC) in Han population in Qingdao. Methods: 225 cases of pathologically confirmed fresh gastric cancer tissues or paraffin embedded gastric cancer tissues during January 2015 and October 2018 were collected from Affiliated Hospital of Qingdao University, as GC group; in the meanwhile, 200 healthy people underwent physical examination at Outpatient were collected as control. The 225 cases of cancer tissues were assigned into two groups according to the transcription result of EBV-encoded small molecule non-polyadenylation (EBER 1) by In situ hybridization: EBVaGC group (70 cases) and EBVnGC group (155 cases). DNA was extracted from the tissues of two groups and peripheral blood of healthy participants.According to the general setting of HaploView software (MAF>0.05, r2>0.8), four TagSNPs (rs217727, rs2735971, rs2839698 and rs3741216) of H19 were screened. Genotyping of each SNP locus was carried out by using Taq-Man MGB allele typing kit, and the gene polymorphisms were examined. Results: H19 SNPs of collected tissue samples were in accordance with the HardyWeinberg equilibrium. Compared with control group, patients carrying TT genotype of H19 rs217727 loci had significantly increased susceptibility to gastric cancer (χ2=9.073, P=0.003, OR=1.999, 95% CI=1.271-3.143), so did the T allele (χ2=13.475, P=0.001, OR= 1.661, 95% CI=1.266-2.180). In contrast, patients carrying TC and CC genotype of rs2839698 loci had significantly increased susceptibility to gastric cancer (χ2=9.407, P=0.002; χ2=6.517, P=0.011), and patients carrying C allele had obviously increased susceptibility to GC (χ2=6.163, P=0.013, OR=1.417, 95%CI=1.076-1.867; χ2=9.542, P=0.02, OR=2.070, 95%CI=1.298-3.302).As for the rs2735971 and rs3741216, there was no significant difference between the GC group and the control group (all P>0.05). The distribution of 4 H19 SNPs showed no statistical difference in both EBVaGC group and EBVnGC group (all P>0.05). Conclusion: There was an association between H19 gene polymorphism (rs217727 and rs2839698) and gastric carcinoma; patients carrying the TT genotype of rs217727 and C allele of rs2839698 may increase the risk of gastric carcinoma. No significant association was observed between H19 SNP polymorphism and risk of EBVaGC.

@#To investigate the iron deficiency (ID) in children with congenital heart disease (CHD) and find high risk factors of ID. Methods    The clinical data of 227 pediatric patients with CHD from February to June 2016 were retrospectively analyzed. The incidence of ID according to the result of iron metabolism examination (serum ferritin <12 μg/L as the diagnostic criteria) was investigated. According to their basic CHD types, patients were divided into a cyanotic group and an acyanotic group. We tried to find the high risk factors of ID in those pediatric CHD patients by comparing their age, gender, growth condition and blood routine test results. Results    There were 19.8% pediatric CHD patients complicated by ID. The incidence of ID in the cyanotic patients was higher than that in the acyanotic patients (31.0% vs. 17.3%, P=0.045). In both groups, ID patients presented the characteristics of younger age, higher anemia rate, lower mean corpuscular volume (MCV), lower mean corpuscular hemoglobin (MCH), lower mean corpuscular-hemoglobin concentration (MCHC) and longer red blood cell distribution width (RDW). Conclusion    Cyanosis, younger age (infant), anemia, decreased MCV, decreased MCH, decreased MCHC and increased RDW are high risk factors of ID in CHD children.
